How to troubleshoot unexpected Linear DNA migration on agarose gels
If your ENFINIA™ Linear DNA appears to run like a longer fragment on an agarose gel, this can be due to sequence adapters or DNA structure.
- ENFINIA™ Linear DNA contains:
- 5’ adapter: 46 bp
- 3’ adapter: 42 bp
- Total added length to your gene of interest: 88 bp
- Additional considerations: Secondary structures in the DNA can cause it to migrate more slowly in non-denaturing gels, making it appear larger than expected.
Recommendations:
- Account for the 88 bp of adapter sequence when comparing to size markers.
- For more accurate sizing, use denaturing gel conditions.
