How to troubleshoot unexpected Linear DNA migration on agarose gels

If your ENFINIA™ Linear DNA appears to run like a longer fragment on an agarose gel, this can be due to sequence adapters or DNA structure.

  • ENFINIA™ Linear DNA contains:
    • 5’ adapter: 46 bp
    • 3’ adapter: 42 bp
  • Total added length to your gene of interest: 88 bp
  • Additional considerations: Secondary structures in the DNA can cause it to migrate more slowly in non-denaturing gels, making it appear larger than expected.

Recommendations:

  • Account for the 88 bp of adapter sequence when comparing to size markers.
  • For more accurate sizing, use denaturing gel conditions.
